If I put a pressure tester on my rad at 16 lbs. " which my pressure is supposed to be", then leave it over night, should it still read 16 Lbs. in the morning "assuming nothing is wrong with the tester"?....thanx in advance....

It won't.
=============== Yes but, a few minutes is all you should need to find the leak.
not for long, in order to check the pressure it ends up using some.
