Not necessarily. There are Welch plugs ('freeze plugs') on the sides of the block. These plug holes which were used to remove the casting sand when the block was poured, and really have nothing to do with freezing temperatures.
These plugs often corrode, leading to coolant leaks. In most cases they are easy to replace.
There may also be Welch plugs in the heads and at the front and back of the engine block.
If you have a failed plug at the back of the block, you will have to pull the transmission to get to it.
