The 2-sided key only works one way, and that way it meets with a stiffness while turning it.
Can I free up the lock by spraying with WD 40 or some other lubricant?

Yes.
Don't use WD40. Oil based lubricants attract and hold dirt and over time tend to make the situation worse. Use a graphite lubricant designed specifically for locks. Available from Autozone, etc. Comes in a "puffer" tube for a dollar or so.

Odds are you've worn the keys (probably copies?). If you have a master key, try it. If you have the master key code, try having a new key made from it, not a copy.
This is probably why one side works, it's either a better copy or not as worn...
If you don't have either, you may need to get a new set of locks/keys.
