I have a 2000 Toyota Avalon with about 274000 Km on the clock and it's developed a knock in the motor but doesn't sound like bearings to me Any ideas what it might be?

We owned a Mazda MPV with the 3.0L V6 that developed an issue with the cylinder barrels going oblong and this caused the pistons to slap the insides of the cylinder walls.
I don't know if Toyota motors have this issue, but it's one of the things to consider looking into.
