Can someone please tell me how to manually tune the radio? The user guide says to press the up/down buttons but this just skips through the stations in memory. Pressing and holding sets it scanning.

They can be funny sometimes. Try it with RDS turned off (just press the RDS button once). Once you have found the channel you want hold the relevant preset button down for 5 seconds until it beeps. You can then turn RDS back on and save it a second time.
